Abstract
THE ULTRASTRUCTURAL HISTOPATHOLOGY OF EXSEROHILUM PEDICELLATUM
(HENRY) LEONARD & SUGGS DURING INFECTION OF MAIZE ROOTS.
The aim of this study has been to determine the interaction between
the fungus pathogen and the host plant on an ultrastructural
level. This pathogen is part of the root rot complex of
maize.
Heat treated and surface sterilized seeds were planted in
pathogen infested vermiculite in pots and also in custom made
root inoculation devices. In addition to TEM, SEM and light
microscopy , fluorescence microscopy as well as a histochemical
procedure for the location of cellulase in infected root tissue,
were employed to study the effect of root infection by the
pathogen. The maize roots were , apart from microscopical techniques,
exposed to an extracted fungus metabolite and commercial
cellulase and pectinase in order to obtain more knowledge about
the pathogen-host interaction. ...
