Abstract
The primary aim of this study was to investigate the electrophysiological mechanisms of toxins (PBTx1, Toxin 1, and Toxin 2) isolated from the venom of the scorpion Parabuthus transvaalicus
. The research compared the effects of Toxin 1 and 2 on cloned human heart sodium channels (hH1) with native channels in rat heart to determine if animal models could accurately reflect human responses
. Key findings showed that native rat heart channels are effective for studying toxin effects
. The study identified a specific α-effect induced by Toxin 1 and 2, which arises from two new toxin structures that are smaller than classic sodium channel toxins
. A kinetic model was developed showing channels moving in parallel pathways, where the toxins exert negative cooperative effects on transitions to the open state and concentration-dependent effects on inactivation
. Both the potassium and sodium toxins were classified into new subfamilies due to their unique structures
